Source text - English Pathogenic fungi have a devastating impact on human nutrition and health. Our group aims to unravel the molecular and genetic bases of fungal pathogenicity. Our working model is Fusarium oxysporum, a soilborne pathogen causing vascular wilt disease on more than a hundred plant species, as well as systemic infections in immunocompromised humans.
We have established a unique multihost infection model based on a single isolate of F. oxysporum f. sp. lycopersici, which is able to cause disease on tomato plants as well as onimmunodepressed mice and the non-vertebrate host Galleria. Targeted mutants in genes of interest can thus be assayed directly for virulence on plant and animal hosts. The availability of the complete genome sequence allows the application of genomic and proteomic approaches in addition to forward and reverse genetics. Using a variety of experimental strategies, our group has identified novel virulence determinants such as signalling components, transcription factors, cell wall remodelling enzymes and secreted proteins.
Our long-term goal is to gain insight into the genetic setup that enables fungal pathogens to cause disease on a wide range of hosts across different eukaryotic kingdoms.

Nossa equipe tem como objetivo esclarecer as bases moleculares e genéticas da patogenicidade fúngica. Nosso modelo de estudo é o Fusarium oxysporum, um patógeno de solo causador da doença da murcha vascular em centenas de espécies de plantas, assim como infecções sistêmicas em humanos imunocomprometidos. Nós estabelecemos um modelo único de infecção com hospedeiros múltiplos baseado numa cepa isolada do F. oxysporum f. sp. lycopersic, capaz de causar doenças em tomateiros, em camundongos imunodepressivos e nos hospedeiros invertebrados Galleria. Assim, a virulência nos hospedeiros plantas e animais podem ser analisadas diretamente pelas mutações dirigidas nos genes de interesse. A disponibilidade da sequência completa do genoma permite a aplicação da abordagem genômica e proteômica, além da genética direta e reversa. Utilizando uma variedade de estratégias experimentais, identificamos novos determinantes de virulência como componentes de sinalização, fatores de transcrição, enzimas remodeladoras da parede celular e proteínas secretadas.
Nosso objetivo a longo prazo é obter um esclarecimento acerca da constituição genética que permitem aos patógenos fúngicos causarem doenças em uma ampla gama de hospedeiros de diferentes reinos eucarióticos.
