Scope:
This document provides information about a Distribution Chain Audit.
Purpose of Distribution Chain Audit
During audits, a team of experts follows the distribution chain making qualitative and quantitative inspections at every stage.
The collected data give information about:
- Type of distribution chain and physical hazards
- Package and secondary packaging appearance at different stages through the chain.
Collected information are used:
- To address customer issues and concerns regarding package performance
- To create recommendations for performance improvement of the audited product

Distribution Chain Stages
Damage on package can occur on each stage of the distribution chain before reaching consumers.
At every stage of DC there are hazards that can cause damages to packages.
Every DC stage can result in different damages on packages depending on the specific actions performed during it.

3.2.1. Thoroughly spray syringes containing lipo-aspirate with 70% ETOH. Cap with syringe tip caps, place tip down on rack inside LFBSC.
3.2.2. Transfer materials listed on the “Materials” table above to the LFBSC. Materials that are not in sterile condition are to be sprayed with 70% ETOH before entering the LFBSC. Label the 250 ml fluid container as “Waste” before transferring to the LFBSC. Record time
3.2.3. Washing the adipose tissue
3.2.3.1. Gently discard the tumescent fluid from the 60cc LL syringes, retain the adipose tissue.
3.2.3.2. Re-distribute adipose tissue among 60cc LL syringes to maintain equal amounts on each syringe. No more than 30cc of adipose tissue on each 60cc LL syringe
3.2.3.3. Aspirate NSS 0.9% from the 1L bag into the 60ccLL syringes as needed to fill completely. Gently turn syringe to wash remaining tumescent fluid out of adipose tissue, allow resting for two minutes. Drain fluid, retain adipose tissue.
3.2.3.4. Repeat step 3 until fluid in the 60cc LL syringes is clear.
3.2.4. Dissociation of adipose tissue
3.2.4.1. Gently discard fluid from 60cc LL syringes, retain adipose tissue.
3.2.4.2. Verify that each syringe has approximately equal amounts of adipose tissue, maximum 30ml.
3.2.4.3. Using the 10cc LL syringe and the 18GX1” needle aspirate the cell extraction medium from the vial and add to the adipose tissue on each syringe. Gently mix syringe content by turning. Record time: _____________AM/PM.
3.2.4.4. Using an empty 60cc LL syringe and a LL to LL connector, gently transfer content from one 60cc syringe to another about 10 times to mix homogeneously.
3.2.4.5. After mixing is completed replenish 60cc syringe with NSS 0.9% until the 45 cc marking.
3.2.4.6. Transfer 60cc LL syringe content to 50cc centrifuge tube.
3.2.4.7. Repeat steps 3 to 6 above for each 60cc syringe until all the adipose tissue is mixed with the extraction medium and stored in 50cc centrifuge tubes.
3.2.4.8. Gently turn 50cc centrifuge tubes for 10 seconds every 5 minutes for 30 minutes after
the time recorded on step 3 above.
3.2.4.9. Centrifuge 50 cc tubes at 1000xG for 10 minutes.

3.2.3.3. Aspire NSS 0,9% de la bolsa de 1L y llene completamente las jeringas LL de 60cc. Gire suavemente la jeringa para quitar el líquido tumescente restante fuera del tejido adiposo, deje reposar por dos minutos. Drene el líquido, conserve el tejido adiposo.
3.2.3.4. Repita el paso 3 hasta que el fluido en las jeringas LL de 60cc esté claro.
3.2.4. Disociación del tejido adiposo
3.2.4.1. Deseche suavemente el líquido de las jeringas LL de 60cc, conservando el tejido adiposo.
3.2.4.2. Verifique que cada jeringa tenga cantidades aproximadamente iguales de tejido adiposo, máximo 30ml.
3.2.4.3. Usando la jeringa LL de 10cc y la aguja 18GX1”, aspire el medio de extracción de células del frasco y añada el tejido adiposo en cada jeringa. Mezcle suavemente el contenido de la jeringa agitándola. Registro de la hora: _____________AM/PM.
3.2.4.4. Usando una jeringa LL vacía de 60cc y un conector de LL a LL, transfiera suavemente el contenido de una jeringa 60cc a otra alrededor de 10 veces para lograr una mezcla homogénea.
3.2.4.5. Después de mezclar completamente se termina de llenar la jeringa 60cc con NSS 0,9% hasta 45 centímetros cúbicos marcados.
3.2.4.6. Transfiera el contenido de la jeringa LL de 60cc al tubo de centrifugado de 50cc.
3.2.4.7. Repita los pasos 3 a 6 mencionados anteriormente para cada jeringa de 60cc, hasta que todo el tejido adiposo se mezcle con el medio de extracción y almacene en tubos de centrifugado de 50cc.
